Loratadine is a drug which is used to in the treatment of seasonal allergic rhinitis. This drug is similar to cyproheptadine and azatadine in structure.Loratadine is basically a non-sedating blocker of H1.According to the tests made Loratidine does not penetrate through CNS.It also has low affinity for H1 receptors. The drug has a weak affinity not only for acetylcholine but also for alpha adrenergic receptors. Loratadine is similar in mechanism with other H1 blockers. It also competes with free histamine.

The reason for this is its binding with H1 receptors. This is done in the GI tract, uterus, muscle particularly bronchial or the large blood vessels.Loratadine has a rapid metabolism of first pass hepatic. It is also absorbed from the body organ or the gastrointestinal tract. This drug is bound to the plasma proteins in the body.Descarboethoxyloratadine or desloratadine is its metabolite which is active. It binds moderately to the plasma proteins.

The effect of this drug is seen in about one or two hour. The biological half life of this drug is about eight hours, while its metabolite’s half life is about twenty eight hours. Some traces of loratadine which is unmetabolised are found in the urine.Loratadine when taken with certain other medications show various drug interactions. The basic function of the drug is to block the action of the histamine receptors. This in turn prevents or reduces the reactions of the immune system which plays the role of causing the allergy symptoms.
